I got this in Pharmacology (Katzung):
The mechanism of toxicity requires oxidation to Cytotoxic intermidates by p450 enzymes.
This occurs if substrates for phaseII conjugation reactions (acetate & glucuronide) are lacking.
The intermidate (Ac) is conjugated with glutathion to harmless product .
If glutathion is exhausted Ac combines with hepatic cell proteins, resulting in cell death.
